EFFICACY OF DIFFERENT DOSES OF ONDANSETRON IN CANCER PATIENTS TREATED WITH CISPLATIN-CONTAINING CHEMOTHERAPY

Background: The aim was to compare the efficacy of different doses of ondansetron (OND) for prevention of cisplatin-induced acute emesis. Methods: A total of 35 untreated cancer patients entered this study. In consecutive courses all patients received chemotherapy for the control of acute emesis (day 1) OND (8 mg i.v. tid) and (OND 4 mg i.v. bid). The patient characteristics were well matched with respect to the gender, weight, age and BSA. Results: Patients were classified as responders and non-responders. Responders were patients with overall response (OR) for vomiting and nausea. Non-responders were patients with minor response + failure for the vomiting and moderate + severe nausea. There was no significant difference in the OR, where 82.9% had OR with OND 24 mg/d and 68.6% with OND 8 mg/d (p>10). Concerning the control of acute vomiting, non-responders were 17.1% in the group treated with OND 24 mg/d and 31.4% with OND 8 mg/d. The same results were obtained in the control of acute nausea. OR have been achieved in 85.7% from the first group and in 74.3% from the second group (p>0.10). Concerning the control of acute nausea, non-responders were 14.3% in the group treated with OND 24 mg/d and 25.7% with OND 8 mg/d. There was significant difference between responders and non-responders concerning the vomiting and nausea. There were not significant differences between non- responders for both grousp. "Rescue" therapy with metoclopramide was used only in one patient, treated with OND 8 mg/d. Conclusion: Our results confirm the opinion about the lack of gemeral consent concerning the most effective dose OND in control of acute emesis. |
