ALTERED OXIDATIVE SUSCEPTIBILITY AND ANTIOXIDATIVE CAPACITY OF ADJACENT HUMAN COLON CARCINOMA TISSUE AND POLYP: THE CAUSE OR CONSEQUENCE OF INCREASED PROLIFERATIVE POTENTIAL

Background: Altered cellular pro-oxidant status as well as decreased antioxidative defense can initiate cells to neoplastic growth. There is evidence that numerous chronic inflammatory gastrointestinal diseases, followed by phagocyte-generated reactive oxygen species, are considered as precanceroses. Methods: The study included 8 patients with colon carcinoma and 6 with polyps. Bioptates were from tissue surrounding malignant lesion and polyp tissue as well as from normal tissue further from alliterative spot. Oxidative susceptibility of bioptate was as from normal tissue further from alliterative spot. Oxidative susceptibility of bioptate was carried out at pro-oxidant conditions (Fe++/ ascorbic acid) to determine the ability of the tissue homogenate to generate additional peroxides. The antioxidative capacity of bioptate was tested using model of Fe++/ascorbic acid - induced generation of MDA in phospholipid liposome suspension and expressed as percent of inhibition of MDA formation. Results: Obtained results show that used pro-oxidant system (Fe++/ascorbic acid) was capable to produce much greater effect in control healthy tissue (MDA concentration 4.14±0.9 and 3.37±0.88) than in corresponding tissue surrounding carcinoma (2.19±9.45; p`0.005 or polyps (1.87±0.59; p`0.01). The antioxidative capacity was increased in tissue surrounding carcinoma 72.2±11.5%) vs. control healthy further tissue (50.7±11.47%; p`0.05). However, there was not significant change in antioxidative capacity of polyp bioptate compared with normal mucous. Conclusion: The obtained results point out the changes in pro/antioxidative status in adjacent human colon carcinoma tissue as well as in polyp, which could indicate the need for more radical surgical intervention. |
